Lemborexant and Daridorexant for the Treatment of Insomnia: An Indirect Comparison, Using Number Needed to Treat, Number Needed to Harm, and Likelihood to Be Helped or Harmed

Abstract Background To describe lemborexant and daridorexant for insomnia treatment using number needed to treat (NNT), harm (NNH), likelihood be helped or harmed (LHH). Methods Dichotomous outcomes were identified from the two registrational randomized controlled trials. Analogous data then extracted registration studies. Results Using pooled dosage data, had a clinically relevant magnitude of therapeutic effect, as evidenced by NNT values versus placebo <10, NNH >10, suggesting that is relatively tolerable. When comparing response discontinuation because an adverse event (AE) Month 3, LHH ranges 5.2 10.4 5 mg 10 doses (a favorable result). For daridorexant, efficacy 25 50 generally result in ≥10; all instances, yield more robust estimates than dose. The rate AE at 3 was higher rendering calculations despite less estimates, with range 90.9 125 when Conclusions Benefit-risk ratio measured NNT, NNH, LHH. Indirect comparisons suggest advantage tolerability daridorexant. Funding Eisai Inc.
